MeCP2 is a protein associated with a variety of neurological disorders, including Rett Syndrome, the most physically disabling of the autism spectrum disorders. The researchers show that MeCP2-deficient astrocytes (a subset of glia) stunt the growth of neighboring neurons. Remarkably, these neurons can recover when exposed to normal glia in culture. This research suggests a novel approach to treating Rett Syndrome.

The researchers show that MeCP2-deficient astrocytes (a subset of glia) stunt the growth of neighboring neurons. Remarkably, these neurons can recover when exposed to normal glia in culture.

The discovery was made by Gail Mandel, Ph. D. of the Vollum Institute at Oregon Health and Science University and a Howard Hughes Medical Institute investigator and Nurit Ballas, Ph. D., a research associate, at the time, in the Mandel lab at the State University of New York at Stony Brook. Dr. Mandel is a scientific advisor of the Rett Syndrome Research Trust (RSRT) a recently formed nonprofit organization intensively focused on the development of treatments and cures for Rett Syndrome and related MECP2 disorders.

Rett Syndrome strikes little girls almost exclusively, with first symptoms usually appearing before the age of 18 months. These children lose speech, motor control and functional hand use, and many suffer from seizures, orthopedic and severe digestive problems, breathing and other autonomic impairments. Most live into adulthood, and require total, round the clock care. Individuals with Rett and their families suffer the emotional and financial cost of the wide range of symptoms and the ongoing struggles to address them.

Glial cells, which reside throughout the nervous system, comprise the vast majority (90%) of cells in the brain. Glia support and interact with neurons in innumerable ways, from providing the structural underpinnings and guidance of axons and dendrites (the neuronal processes that carry information), to creating protective insulation for axons. These and other glial functions are essential to the health of neurons.

Since MECP2 is located on the X chromosome, girls with Rett Syndrome are mosaic: cells with a healthy copy of the gene lie side by side with cells that have the mutated version. Mandel and colleagues found that MECP2- deficient astrocytes could not sustain normal neuronal growth. In fact, their experiments suggest that MeCP2-deficient astrocytes secrete a toxic factor that impacts the entire neighborhood of neurons, including neurons with wildtype (normal) MECP2.

Conversely, they found that wildtype astrocytes could restore the stunted growth of MeCP2-deficient neurons. This finding supports the 2007 Science publication by Professor Adrian Bird showing reversal of Rett symptoms in adult mouse models of the disorder. Results from ongoing in vivo mouse studies deleting MeCP2 in glia are promising in terms of supporting the culture studies.

"This new study adds to the growing body of evidence that glial cells are critically important contributors to neurological diseases and therefore attractive targets for drug development," said Ben Barres, Professor and Chair of the Department of Neurobiology at Stanford University and a glial cell expert.

Recent studies from multiple groups, including the lab of Dr. Tom Maniatis, a neurodegenerative disease researcher at Harvard University, have implicated glia in Lou Gehrig's disease (Amyotrophic Lateral Sclerosis - ALS), a devastating neurodegenerative disorder that leads to the death of motor neurons and total paralysis. "In Rett Syndrome, faulty glia seem to poison neurons, inhibiting growth; in ALS glia appear to release a toxic factor that kills motor neurons." Maniatis stated that "studies of the role of glia in a broad spectrum of neuronal diseases should lead to exciting advances in understanding disease mechanisms."

"Dr. Mandel's sharp insights have given the scientific community an entirely new way to think about treating Rett Syndrome. One could envision a scenario where halting the secretion of this potential toxic factor could restore health, in particular to the MECP2 positive neurons, which could bring about amelioration of symptoms. I look forward to her lab's continued contributions as we explore the therapeutic implications of her research," comments Monica Coenraads, Executive Director of RSRT and parent of a child with the disorder.

Other people who worked on the project include Daniel T. Lioy of the Vollum Institute and Christopher Grunseich, M. D. while at the State University of New York at Stony Brook. Nurit Ballas is now a Research Associate Professor in the Department of Biochemistry and Cell Biology at Stony Brook.

The MISSing Angels Bill helps provide much-needed comfort, dignity, and documentation to women and their families experiencing the death of a baby just prior to or during birth. All states, including New Mexico, require the family to pay for funeral expenses, and a death certificate/report is issued after a stillbirth. Yet, some states refuse to offer women the choice of a birth certificate. It offers an option to women who give birth to loved and wanted babies who die unexpectedly to choose a birth certificate.

"I would assume that the last thing a presidential candidate would want to do is knowingly wound thousands of grieving mothers after their baby's death," said Daryl Logullo, Legislative Liaison for the MISS Foundation.

In response to the veto, the MISS Foundation has released a public service announcement that portrays the experiences of bereaved parents after stillbirth and is directed at Richardson's campaign. A very sensitive and controversial video, it speaks of the enormity of this public health issue. "Bill Richardson made a grave error in judgment, clearly being misinformed about this issue by his advisers," said Joanne Cacciatore, CEO of the MISS Foundation. Both the MISS Foundation and the National Stillbirth Society are calling for Governor Bill Richardson to offer a public apology to these bereaved mothers and families.

Bells will be ringing in the homes of Scottsdale residents Ethel and Keith Kolerus this holiday season. The mother-son duo, along with the rest of the family, has made bell-collecting part of a treasured family tradition.

The practice began in 1943 when Ethel, as a new bride, first visited her husband, Frank, who was serving in the army at a base in West Palm Beach, Fla. To commemorate the occasion, Ethel purchased a decorative bell from a nearby gift store to remind her of the event.

Soon collecting bells became a family hobby. When Ethel and Frank traveled, they added new bells to their collection. When children, Keith and Nancy came along, they joined in the fun, often marking holidays and special occasions with gifts of unique bells.

Locating hard-to-find bells became a challenge for family members and friends, and became an important part of family vacations as they traveled to new locations.

Today, the collection consists of over 300 bells in a variety of shapes, sizes, materials and styles from all parts of the globe. The assortment includes a complete set of Waterford crystal bells, a small brass bell from Holland, a bell from VietNam and several delicate china bells shaped like Elizabethan-era dolls wearing hooped skirts.

Other distinctive ringers include a small hand-carved teakwood bell from Indonesia, a brass bell sent by a friend stationed with the Strategic Air Command in Africa and a large gong from India topped with miniature ivory elephants.

The collection also features a Dresden bell that is a copy of the Freedom Bell of West Berlin, a bell with a stunning inlaid jade handle, a hand-wrought silver bell and a wall-mounted San Francisco trolley bell, a gift from EthelÂ’s son, Keith.

Also included in the collection is a large number of animal bells used to track flocks of camel, goats, turkeys and elephants. A special elephant bell purchased during a 1958 vacation to Niagra Falls was the mate to one used by a delegate at the National Republican Convention that year.

Other bells have interesting histories. For example, a series of brass gongs in graduated sizes was sent to Ethel by a friend who served as a missionary in China. There, the bells were rung to announce services or call people together for special events.

Because Ethel believes the collection should be used as much as possible, she has loaned a number of bells to deserving groups over the years. The Chinese mission gongs, for example, were used as props in her daughter, Nancy’s, Mt. Prospect High School production of “The Good Earth.”

Other bells were lent to Girl Scout troops studying the customs of foreign countries, and Ethel, former secretary of IllnoisÂ’ Northwest Suburban Chapter of the American Cancer Society, loaned a number of smaller bells to bedridden cancer patients who used them to summon family members or healthcare workers to their bedsides.

One of the most prized and nostalgic bells in her collection is a large bell that hung in the bell tower of the Grimms Public School in Grimms, Wis., where her husband Frank, now deceased, attended school.

For over 98 years, the bell summoned children in the community of 68 to a one-room schoolhouse. Today, the bell sits in a cupola atop Keith KolerusÂ’ new 9,000-square-foot home on Pinnacle Peak.

Much of the collection is now kept in boxes at KeithÂ’s home, but some of it remains on display in EthelÂ’s apartment at Sierra Pointe retirement community. As she moves among the familiar bells, chimes and gongs, tapping one occasionally to take in its clear resonance, she smiles as if greeting an old friend.

Ethel believes her hobby has brought her and the rest of the Kolerus family a great deal of pleasure over the years. Looking at the bells, she says, is like visiting old friends and relatives. Each evokes memories of some special place or time.

During the holidays, Ethel, Keith and other family members will ring the old school bell to commemorate a family tradition of tintinnabulation that has lasted over 61 years.

Technology seen as key enabler to improve tax effectiveness

The research report titled Improving Corporate Tax Effectiveness: Trends and Technology was prepared by CFO Research Services and underwritten by Longview Solutions. The report is based on a survey of 190 senior tax executives at companies with annual revenues ranging from US$1 billion to more than US$20 billion in the United States, Canada, and Europe and examines their views on tax function effectiveness, their strategic priorities, and their future outlooks.

Key findings of the research include:
 Over 80% of respondents are somewhat or much more focused on improving profitability or maintaining cash flow. At the same time, more aggressive enforcement and pressure to increase reporting transparency will continue to test the tax function’s resources in the coming months.  Though under-resourced, many tax departments plan to pursue a wide range of improvements to promote productivity and boost efficiency amid complexity.  Forty-four percent of respondents foresee an important role for technology improvement and an additional 32% confirm it will play a critical role in efforts to improve tax effectiveness.  Standardization and integration of data for tax and financial reporting purposes are a top priority on the tax function’s information agenda.  A substantial number of respondents who agree that it's important for their companies to document the business case for tax technology also say their companies are not well-equipped to document various dimensions of that business case. For example, 40% of respondents who agree that it's important to quantify the return on tax automation also say their companies are not well-equipped to document that aspect of the business case for investment.

As they brace themselves to meet a countless number of new requirements, including increasing pressure for transparency in tax reporting, new (and swiftly-changing) tax legislation, and increasingly aggressive enforcement, many executives responding to the survey seem to place a premium on flexibility. “We are in a new and dynamic period of change–with an almost certain raft of changes in regulatory and tax generation legislation,” one respondent writes in response to an open-ended question. “Being as automated as possible minimizes 11th-hour firefighting by cutting down on human interventions,” the respondent continues.

But the research also confirms that many tax departments have been seriously under-resourced both in terms of human capital and technology through the course of the economic downturn, says Celina Rogers, Director of Research at CFO Research Services. “Regulatory, legislative, and enforcement pressures are ratcheting upward, and tax executives expect those trends to continue,” says Ms. Rogers. “It's also clear from the research that tax executives see the value in tax automation and, more particularly, in integrating processes and systems for financial and tax reporting, as they work to meet those challenges.” But, Ms. Rogers points out, survey results showing some uncertainty among respondents about their ability to make the business case for tax technology investment suggest that vital technology initiatives may go unfunded at many companies. “Building the business case for investment is always important, but in an environment of extremely scarce resources and uncertainty about the strength of the recovery, the ability to convincingly document the business case for investment is critical. Tax departments that aren't able to do so are likely to see technology and other improvement initiatives go unfunded,” she says.

“As a thought leader in the area of tax technology, we feel it is important to conduct third party research to ensure we have a clear understanding of the challenges and opportunities our clients face day in and day out,” said John Power, President, Longview Solutions. “These tax executives are struggling to help their companies improve profitability without the tools and automation necessary to help with the heavy lifting. According to a recent survey, fewer than 20 percent of Americans can state their credit score within an appropriate range -- and almost half of respondents never check their credit score -- which indicates the time is right for a credit checkup, according to Andrew Housser, co-founder and co-CEO of online financial portal Bills. com.

"Around the holidays, we tend to think of spending, not saving. But the holidays are a great time to take a look at your personal fiscal status," Housser said. He pointed out that for those in danger of holiday overspending, a December review might help rein in expenses. Additionally, a review now will prepare consumers for next year's tax season, and allow them to make any necessary changes to savings plans and payroll deductions for 2007.

Individuals have most of the information they need to understand their financial picture, and other information is readily available, Housser explained. "Best of all, conducting an annual financial examination is even easier than scheduling an annual physical exam -- and less painful."

Here are Housser's steps to checking one's own financial picture:

1. Free credit score gives a baseline. An individual's credit score determines the type and amount of credit for which he or she is eligible, as well as the interest rate. While the survey showed many Americans are ignorant of their creditworthiness, about one-third are on the right track -- checking their credit score annually. Request a free credit report once every 12 months by visiting www. annualcreditreport. com or calling 1-877-322-8228.

2. Net worth provides the big picture. "Net worth" is the value of assets minus the value of debts. Add cash on hand, savings and checking accounts, retirement accounts and the value of property such as a home and automobiles. Then add up debt on credit cards, mortgages, auto loans, student loans and other debt. "Subtract what you owe from what you own. The result is your net worth," said Housser, who pointed out that for those with a lot of debt, the net worth might be negative.

3. Take a savings temperature. In 2005, Americans had a negative savings rate -- spending more than they earned -- for the first time since the Great Depression. Calculate a savings rate by adding up all savings -- including retirement contributions and employer matching, health savings account contributions and even spare change on its way to the bank. To find savings as a percentage of disposable income, take the net (after-tax or take-home) income and subtract mandatory expenses (rent, utilities, basic food expenses). The remainder is the disposable income. Then, divide monthly savings by monthly disposable income to find the savings percentage. "Aim for at least 10 percent going into a savings vehicle," Housser advised.

4. Check credit card payoff timelines. Payments on credit card debt are like a gift to the bank -- especially in the case of minimum payments. Gather credit card statements and use an online calculator (search "credit card payoff calculator") to determine the estimated payoff date for each card. "Then plan to pay them off using a tried-and-true method," Housser said. "First, stop charging. Second, pay as much as you can each month on the highest-interest debt, while making minimum payments on other cards. Third, when you've paid off the highest-interest card, add that payment amount to your minimum payment on the second-highest-rate card until that debt is repaid. Continue until your debts are gone. Then, you can start saving -- paying yourself! -- what you've been giving to the bank."

Whatever your financial dreams, the first step to achieve them is understanding where you stand. By checking your financial position each year, you can decide your next move -- and that's a very healthy plan."

In immediate response to the Muscular Dystrophy Association's decision to cancel the remainder of its annual summer camp program due to the H1NH virus, more commonly known as the "swine flu" Camp Promise, today announced its efforts to provide an alternative to those children who have already signed up and for those that would like to sign up to attend.

Camp Promise, based out Bedford, NH is lead by volunteers, parents and supervised by accredited physicians. The health, safety and general well-being of these children is taken very seriously. Camp Promise follows all protocols to prevent the spread of any and all potentially infectious disease. All campers, staff and volunteers will be screened before and during the camp session.

"As parents of children with neuromuscular disorders we possess a heightened awareness of all potential risks that may endanger our children," said Natalie Gaudenzi, parent and advocate of Camp Promise. "Myself -- along with 18 other parents -- have given careful consideration to this threat and believe that the reward for our children as well as the entire family is far greater than risk of swine flu or that of any other risk we encounter in our daily lives. Many of these children have been looking forward to this experience for a long time and some-due to age restrictions-may never have this opportunity again."

Camp promise will be held at Camp Allen in Bedford, New Hampshire from July 5 through July 10, 2009 and would like parents to be aware that there is a risk of contracting the virus, however, the camp staff and medical staff at Camp Promise will undertake every precaution to prevent the start and spread of the virus.

Camp Promise will begin raising money today to help organize and facilitate an alternate campsite for children between the ages of 7 and 17 who have already signed up for the MDA camp and will also open up its doors to those whose children were not signed up and would like to attend. Camp Promise will offer immediate priority to those children who were attending the MDA camp and will give the remaining open slots on a first come, first serve basis. The cost is approximately $675.00 per child, however Camp Promise is accepting donations to help launch the program and subsidize those children who are not able to meet the price requirements.

North American Firms Trail Europeans for Quality of Investor Relations Websites, Study Finds

When it comes to using the Internet to communicate with shareholders and investors, the average European large-cap company is doing a better job than its North American peers. German companies are the best, Japanese are the worst, and US and Canadian firms seventh and nineth respectively.

Spearheaded by German, Swedish, British and Swiss companies, European investor relations professionals are making better use of the Internet in their IR programs than their counterparts in the United States, Canada and other countries.

This is the main finding of the second semi-annual study of 507 investor relations websites of large-cap international companies for IR Web Report's IR Website Global Rankings™.

Companies' investor relations pages were evaluated for 100 best practice attributes relating to completeness, transparency, responsiveness and usability. The content of sites accounts for 75% of a siteÂ’s total score and covers all financial disclosures, corporate governance, shareholder resources and stock market information. The final 25% of a siteÂ’s score relates to the usability of the site, including navigation, document formats and internationalization.

The findings indicate that European companies generally take an investor-centric approach to their IR websites and are more prone to best practice, while companies in other countries appear to be oriented more to meeting minimum mandatory requirements, but not necessarily presenting information in ways that are easy for investors to access or use.

Geographic differences are clearly evident in the rankings. A wide spread exists in the scores between the top and worst ranking countries. Germany had the best IR websites while Japan had the weakest. US companies ranked seventh overall and Canadian firms ninth. One of the most improved performances was among Italian companies, which may be responding to the sharp focus on their practices in the wake of the Parmalat debacle.

Meanwhile, score ranges were tighter on a sector basis with little difference between the standards of companies in different sectors. Based on median scores, Telecommunications firms ranked the highest and Health Care companies the lowest for the overall effectiveness of their IR websites.

The highest ranked company among the 507 ranked is Stora Enso, a paper products company based in Finland. The highest ranked US company is Verizon Communications, while BCE Inc. was rated as having the best investor relations site in Canada.

Smalls' vision for FC Harlem is to make it the first elite, inner city soccer academy in the United States. It never caught on like basketball or American football," said Smalls. "It will be a journey, but I am convinced that soccer will be a force of social change in urban neighborhoods."

Smalls would like to see the academy take the form of a unique school model that has a dual focus of developing top soccer players and student athletes. The academy will include a training facility and community center, which Smalls hopes to open in 2014. The facility will hold administrative offices, computer suites, classrooms, recreational and training areas, a cafeteria, indoor soccer and futsal fields and a retail/pro shop. This sports complex will be the home of FC Harlem's elite travel clubs and will allow the non-profit to facilitate all its programs on a year-round basis. The club's Harlem location and clientele promise to make this facility the first soccer complex of its kind in the US.
